The developing brain is more sensitive to drugs than the adult brain and exposure during development has been implicated in the pathogenesis of neuropsychiatric and neurological disorders. Zebrafish has emerged as a useful animal model to assess the developmental neurotoxicity (DNT) of drugs. The advantages include economic high fecundity, the ability to absorb a wide range of drugs, high conservation of gene function compared with humans, ease of genetic manipulation, ex utero development and transparency allowing unobstructive observation of morphological changes using fluorescence technology. Zebrafish larvae also exhibit a number of simple and complex behavioral programs. In this study, we assessed the DNT of antithyroid drugs (methimazole and propylthiouracil) and anticonvulsant (valproic acid) using zebrafish based on the genomic, morphological and behavioral changes and were able to show that the systems pharmacology can be useful to elucidate molecular mechanisms underlying DNT of these drugs.
